Role in pathogenesis
PI3K is activated downstream of STAT5 in the KIT D816V signaling cascade. Activates AKT and subsequently mTOR, promoting abnormal mast cell development. BEZ235 (dual PI3K/mTOR blocker) shows growth-inhibitory effects on neoplastic mast cells in preclinical studies.
